چکیده
Project Goals: We are developing Bacillus megaterium as a host for continuous biofuel production coupled with in situ product extraction by supercritical CO2 (scCO2) stripping. We employ a cross-disciplinary collaborative approach to achieve the following: (1) Develop a supercritical CO2 tolerant strain of B. megaterium into a bioproduction host for biofuels (2) Engineer de novo pathways for biosynthesis of longer chain fuels in B. megaterium and (3) Develop and model a two-phase stripping chemostat for continuous biosynthesis and in situ extraction of biofuels using scCO2 as a sustainable extractive solvent. The abstract below reports on progress towards goals (1) and (3).

Metabolic engineering of Pichia pastoris for production of isobutanol and isobutyl acetate
Background Interests in renewable fuels have exploded in recent years as the serious effects of global climate change become apparent. Microbial production of high-energy fuels by economically efficient bioprocesses has emerged as an attractive alternative to the traditional production of transportation fuels. Here, we engineered Pichia pastoris, an industrial workhorse in heterologous enzyme p...

Global warming and decreasing fossil fuel reserves have prompted great interest in the synthesis of advanced biofuels from renewable resources. In an effort to address these concerns, we performed metabolic engineering of the cyanobacterium Synechocystis sp. strain PCC 6803 to develop a strain that can synthesize isobutanol under both autotrophic and mixotrophic conditions. With the expression ...
متن کاملRedesigning Escherichia coli metabolism for anaerobic production of isobutanol.
Fermentation enables the production of reduced metabolites, such as the biofuels ethanol and butanol, from fermentable sugars. This work demonstrates a general approach for designing and constructing a production host that uses a heterologous pathway as an obligately fermentative pathway to produce reduced metabolites, specifically, the biofuel isobutanol. Elementary mode analysis was applied t...
متن کاملCytosolic re-localization and optimization of valine synthesis and catabolism enables inseased isobutanol production with the yeast Saccharomyces cerevisiae
BACKGROUND The branched chain alcohol isobutanol exhibits superior physicochemical properties as an alternative biofuel. The yeast Saccharomyces cerevisiae naturally produces low amounts of isobutanol as a by-product during fermentations, resulting from the catabolism of valine. As S. cerevisiae is widely used in industrial applications and can easily be modified by genetic engineering, this mi...
متن کاملDevelopment of A Novel Gene Expression System for Secretory Production of Heterologous Proteins via the General Secretory (Sec) Pathway in Corynebacterium glutamicum
Background: Corynebacterium glutamicum (C. glutamicum) is a potential host for the secretory production of the heterologous proteins. However, to this date few secretion-type gene expression systems in C. glutamicum have been developed, which limit applications of C. glutamicum in a secretory production of the heterologous proteins. Objectives: In this stu...
